General description

Glutamine methylation occurs on translation termination factors and ribosomal proteins. Protein methylation has been reported to affect various cellular processes including protein stability, protein-protein interaction, and protein localization. Methylation of histone is suggested as a platform to recruit and regulate other chromatin-related factors. Glutamine methylation is brought about by the action of protein glutamine methyltransferase HEMK2 that specifically requires a GQX3R motif for methylation activity. This antibody was tested for glutamine methylation in chromodomain helicase DNA-binding protein 5 (CHD5) that contains multiple PHD and chromo domains and is reported to function as a tumor suppressor.

Specificity

This rabbit polyclonal antibody detects methylglutamine in proteins.

Immunogen

KLH-conjugated Gly-Gln (Me)-Gly tripeptide.

Application

Anti-Methyl-Glutamine, Cat. No. ABS2185, is a rabbit polyclonal antibody that detects glutamine methylated proteins. It has been tested for use in Western Blotting.
Western Blotting Analysis: A 1:3,000 dilution from a representative lot detected Methyl-Glutamine in CHD5 unmethylated & CHD5 methylated recombinant lysate (Coutesy of, Dr. Kusevic, Dr. Kuditipudi and Dr. Jeltsch at University Stuttgart, Germany).

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected Methyl-Glutamine in in-vitro methylated CHD5, NUT, and eRF1 proteins (Kusevic, D., et. al. (2016). J Biol Chem. 291(12):6124-33).

Quality

Evaluated by Western Blotting in methylated recombinant chromodomain helicase DNA-binding 5 (CHD5) protein, but not in unmethylated recombinant CHD5.

Western Blotting Analysis: A 1:3,000 dilution of this antibody detected Methyl-Glutamine in methylated recombinant chromodomain helicase DNA-binding 5 (CHD5) protein, but not in unmethylated recombinant CHD5.

Target description

~51 kDa observed (for methylated CHD5). Uncharacterized bands may be observed in some lysate(s).

Antiserum was depleted by running over unmethylated protein column.
