General description

GAP-43, a common marker of differentiating neurons, is expressed at elevated levels by developing or regenerating neurons during axon growth. While GAP-43 is an integral membrane protein associated with the cytoplasmic surface of axonal growth cones (and synapses), it is absent from dendritic growth cones. In adult brain, GAP-43 is found in high concentration in presynaptic areas where memory formation is thought to occur, such as frontal cortex, limbic system and hippocampus. Phosphorylation of GAP-43 by PKC (at Ser41 on human, mouse, rat, and bovine GAP-43 or Ser42 on chicken and Xenopus GAP-43) is specifically correlated with certain forms of synaptic plasticity.

Specificity

GAP-43 phosphoSer41. The labeling of the GAP-43 protein (~43kDa) in Western blots of rat brain is blocked by the phosphopeptide but not by the dephosphopeptide. In some tissues the antibody may recognize a lower molecular weight band. This band is also detected by a pan GAP-43 antibody.

Immunogen

Epitope: phosphoSer 41
Synthetic peptide corresponding to amino acids surrounding the phosphoSer41 of rat GAP-43. This is the site phosphorylated by protein kinase C.

Physical form

Affinity purified immunoglobulin in 10 mM HEPES (pH 7.5), 150 mM NaCl with 100 μg/mL BSA and 50% glycerol.

Storage and Stability

Maintain at -20°C in undiluted for up to 6 months after date of receipt. Avoid repeated freeze/thaw cycles. Do not store in a self defrosting freezer.
