Eine zellpermeable Nicht-Peptidyl-Thiazolyl-Thiadiazol-Alternative zu pepJIP1 (TI-JIP1_153-163) und TAT-pepJIP1 (Best.-Nr. <;A href="https://./?[islink]ViewProductDetail-BySKU&;#124;&;#124;SKU&;#124;EMD_BIO-420133&;#124;[/islink]">;420133<;/A>; bzw. <;A href="https://./?[islink]ViewProductDetail-BySKU&;#124;&;#124;SKU&;#124;EMD_BIO-420134&;#124;[/islink]">;420134<;/A>;), die als ein auf die JIP-Dockingstelle gerichteter (IC₅₀ = 239 nM gegen die pepJIP1-Binding an JNK1) und mit Substrat konkurrierender JNK-Inhibitor (IC₅₀ = 700 nM gegen die ATF2-Phosphorylierung durch JNK1) fungiert, während wenig Aktivität (IC₅₀ >;50 µ;M) gegen p38α;, Akt, Furin oder den Letalfaktor stattfindet. Hemmt nachweislich die Phosphorylierung von überexprimiertem GFP-c-Jun in HeLa-Kulturen nach der TNF-α;-Stimulation (IC₅₀ = 6,23 µ;M) in-vitro und stellt die Insulinempfindlichkeit bei einem Mausmodell mit Typ-2-Diabetes (25 mg/kg, intraperitoneal) in-vivo wieder her. Eine Biostabilitätsanalyse an Rattenleber-Mikrosomenpräparationen ergab eine Halbwertszeit der Verbindung von 27 Minuten bei 37,5 °;C.
