General description

The protein kinase AtMPK3 is a component of the MAP kinase cascade.

Specificity

Anti-AtMPK3 recognizes AtMPK3.

Immunogen

Synthetic peptide corresponding to amino acids at the C-terminus of Arabidopsis thaliana MPK3. This sequence is specific to AtMPK3 (not found in the other 20 known AtMPK isoforms). It has limited homology (58% identity) to the AtMPK6 isoform and shares significant homology (67% identity, continuous epitope) with the Nicotiana tabacum ortholog WIPK.

Application

Anti-AtMPK3 antibody produced in rabbit has been used in immunoblotting analysis.

Biochem/physiol Actions

AtMPK3 is a component of the plant MAPK signaling cascade that functions downstream of the FLS2 receptor and regulates the innate response to bacterial and fungal infections. AtMPK3 is also involved in the phosphorylation of VIP1 in response to Agrobacterium infection and the subsequent transfer of the bacterial T-DNA into the nucleus. RNAi mediated inhibition of AtMPK6 causes prolonged activation of AtMPK3 . Anti-AtMPK3 antibody is specific for AtMPK3 (43 kDa). The product also reacts with Nicotiana tabacum and p56 in Papaver rhoeas . In immunoblotting, staining of AtMPK3 is specifically inhibited with AtMPK3 immunizing peptide (Arabidopsis thaliana, amino acids 359-370).

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.

Storage and Stability

For continuous use, store at 2-8 °C for up to one month. For extended storage, freeze in working aliquots. Repeated freezing and thawing is not recommended. Storage in frost-free freezers is also not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilutions should be discarded if not used within 12 hours.
