General description

Cell division control protein 45 homolog (UniProt O75419; also known as PORC-PI-1) is encoded by the CDC45 (also known as CDC45L, CDC45L2, UNQ374/PRO710) gene (Gene ID 8318) in human. Recruitment of replication initiation proteins at the G1/S transition enables origin DNA unwinding and loading of DNA polymerases. CDC45 is a replication initiation protein that plays an essential role in the establishment and progression of the replication fork. Cdc45 associates with the heterohexameric Mcm2-7 helicase and the GINS complex (go-ichi-nii-san, Japanese for 5-1-2-3, for the subunits Sld5, Psf1, Psf2 and Pfs3) to form the CMG (Cdc45-Mcm2-7-GINS) complex. CMG represents the eukaryotic replicative DNA helicase that unwinds origins of replication and drives the replication fork. CDC45 is also reported to interact with several other replication proteins, including DNA helicase B (DHB), Mcm10, Orc2, TopPB1, and DNA polymerases alpha, delta and epsilon.

Specificity

This polyclonal antibody detects CDC45 C-terminal region highly conserved among human, mouse, rat, and hamster species.

GST-tagged recombinant human CDC45 C-terminal fragment.

Application

Detect CDC45 using this rabbit polyclonal Anti-CDC45 Antibody, Cat. No. ABC962, validated for use in Immunocytochemistry and Western Blotting.
Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected CDC45 in L929 murine fibroblasts.

Quality

Evaluated by Western Blotting in L929 fibroblast lysate.

Western Blotting Analysis: A 1:5,000 dilution of this antibody detected CDC45 in 10 µg of L929 murine fibroblast lysate.

Target description

~75 kDa observed. 65.57/60.25/68.77 kDa (human isoform 1/2/3), 65.38 kDa (mouse) calculated. Uncharacterized bands may be observed in some lysate(s).
