General description

Osteocalcin (OC/OCN) is a bone-building protein.[1] This γ-carboxylated protein is made up of 49 amino acids and is synthesized by osteoblasts, odontoblasts and hypertrophic chondrocytes.[2] The bone γ-carboxyglutamate protein (BGLAP), that produces OCN is located on human chromosome 1q22.[3]
This ELISA antibody pair detects human Osteocalcin. Other species are not determined.

Application

For research use only. Not for use in diagnostic procedures.
Please refer to the attached Protocolfor details.

Biochem/physiol Actions

Osteocalcin (OC/ OCN) plays a major role in the bone remodeling cycle, affecting bone mineralization. It is considered as a marker of bone formation. OCN guards against the progression of diabetes and the metabolic syndrome.[2]

Questions

1–4 of 4 Questions
  1. Good day, I would like to know what molecule this kit detects, carboxylated osteocalcin or subcarboxylated osteocalcin?

    1 answer
    1. The capture and detection antibodies are mouse monoclonal. The immunogens are recombinant human Osteocalcin with glutathione s-transferase (GST). These detect the large NH2- terminal fragment and intact hOC. The specificity of activated vs. inactivated forms has not been determined.

      Helpful?

  2. Does the RAB1073 Kit detect both activated and inactivated osteocalcin, or just one of them?

    1 answer
    1. The Capture and Detection Antibodies are of the mouse monoclonal type. The immunogens are derived from recombinant human Osteocalcin with glutathione s-transferase. These antibodies are capable of detecting the large NH2-terminal fragment and intact hOC. However, the specificity of activated versus inactivated forms has not been tested.

      Helpful?

  3. Does this Human Osteocalcin Kit measure total osteocalcin level, meaning the sum of undercarboxylated and carboxylated Osteocalcin? or a specific type?

    1 answer
    1. We have not tested this ELISA kit for specificity of undercarboxylated and carboxylated Osteocalcin; here is information about capture and detection antibodies used in the kit:
Capture and Detection Antibodies are mouse monoclonal.
Immunogens: Recombinant human Osteocalcin with glutathione s-transferase.
Detects the large NH2- terminal fragment and intact human osteocalcin.
The specificity of activated vs. inactivated forms is not tested.

      Helpful?

  4. I am writing this message to ask for a detail protocol of RAB1073 (Human Osteocalcin ELISA). I have checked your website and I could not find a detail protocol to process my experiment. Could you please send me one PDF?

    1 answer
    1. The protocol is attached to the lot-specific Certificate of Analysis.
