General description

Deimination (citrullination) is a post translational modification in which the peptidyl-arginine deiminase family of enzymes convert the guanidino side chain of the amino acid arginine into a ureido group, yeilding peptidyl-citrulline. As a result, anti-peptidyl-citrulline antibodies should also cross react with peptidyl-homocitrulline, produced by the carbamylation of lysine, but not free citrulline, which is created in the urea cycle, or as a by-product of nitric oxide synthesis.

Immunogen

Citrullinated peptide consisting of 10 citrulline residues and a carboxyl Gly-Gly-Cys.

Application

Immunohistochemistry Analysis: A 1:250 dilution from a representative lot detected peptidyl-citrulline in human cerebellum, and rat cerebellum tissue.
Western Blotting Analysis: A representative lot detected peptidyl-citrulline in CNS proteins from brains and spinal cord of EAE mice (Nicholas et al., 2005).

The bovine immune system is known for its unusual traits relating to immunoglobulin and antiviral responses. Peptidylarginine deiminases (PADs) are phylogenetically conserved enzymes that cause post-translational deimination, contributing to protein moonlighting in health and disease. Postranslational modification of proteins can lead to the production of autoantibodies and loss of immune tolerance. This process has been hypothesised to be a critical factor in the pathogenesis of rheumatoid arthritis. The objective of this study was to demonstrate
