Application

L'huile minérale est utilisée pour recouvrir des cultures embryonnaires en microgouttes.

Analysis Note

Cette substance est testée en recouvrement de cultures embryonnaires en microgouttes. (Toutes les substances testées sur embryons sont testées sur des embryons de souris.) Des hybrides BCF1 × BDF1 sont généralement utilisés. Cette huile permet surtout d'éviter l'évaporation du milieu, notamment lors de la manipulation des embryons1. Le protocole est le suivant : Des embryons unicellulaires (BCF1 × BDF1) sont mis en culture dans trois microgouttes de milieu HTF (fluide tubaire humain) testé sur embryons supplémenté avec 0,4 % de sérum albumine bovine. Pour que le test soit valide, au moins 70 % des embryons unicellulaires témoins doivent atteindre le stade blastocystaire dans les 96 heures. Pour que le produit soit commercialisable, au moins 80 % des embryons unicellulaires témoins doivent atteindre le stade blastocystaire dans les 96 heures avec la substance testée.

Disclaimer

Ce produit n'est pas stérile. L'huile minérale est difficile à stériliser, hormis par filtration fine de l'huile chauffée. Des antibiotiques sont censés être ajoutés au milieu de culture cellulaire. Si ce n'est pas le cas, utiliser le produit réf. M5310, car il est stérilisé par filtration.

  1. What is the shelf life of this oil?

    1. This product is not assigned an expiration or retest date. Products that are robust in nature and extremely stable, in the original unopened container, are not assigned an expiration date or retest schedule. These products will have no dating reported on either the label or the Certificate of Analysis. Products that are not assigned an expiration or retest date are covered by a warranty period of 1 year from the date of product shipment.

  2. Can M8410 Sigma-Aldrich Mineral oil be used for the deparaffinization of FFPE samples, and is it compatible with downstream histological or molecular analyses like immunohistochemistry or RNA/DNA extraction?

    1. This product has not been validated for deparaffinization of formalin-fixed, paraffin-embedded samples. However, an article in "Applied Immunohistochemistry & Molecular Morphology" (2014 Sep;22(8):623-6) indicated that mineral oil appears to work well for deparaffinizing these samples and for DNA extraction.

  3. Hello,I´m looking for a mineral oil for sulfur pattern production. It´s difficult to find an oil in which the content of sulfur compounds appears in the COA. Can you inform if there´s a possibility of sulfur contamination in the production of mineral oil?

    1. The manufacturing process for this product is considered proprietary. Currently, there is no product option available that is tested for the presence of sulfur.

    1. This material has not been validated in the process of making carbon paste electrodes. However, if the specifications (found below) meet your requirements, then there is no reason that they can't be used in this process.

  6. What is the composition of the oil? Where does it comes from?

    1. M8410 is a light oil. Mineral oil is a complex mixture of different hydrocarbons and its typical composition is as follows:

      Paraffinic oils, based on n-alkanes, 15-60%
      Naphthenic oils, based on cycloalkanes, 30%-60%
      Aromatic oils, based on aromatic hydrocarbons, 3-30%
      Light and heavy mineral oil differ in terms of composition, viscosity, and density.
      Mineral oil with an alkyl chain carbon number of 15 to 25 is categorized as light oil whereas 25 to 50 as heavy oil.
      Light oil has a density of 0.83–0.86 kg L-1 whereas heavy oil has 0.875–0.905 kg L-1
      Light mineral oil is less occlusive than heavy mineral oil as it contains more cyclic (saturated) molecules
