Informations sur le gène

human ... CREB3L3(84699)

Spécificité

This rabbit polyclonal antibody detects CREB-H (CREB3L3). It targets an epitope within the internal region.

Immunogène

GST/His-tagged recombinant fragment corresponding to 92 amino acids from the internal region of human Cyclic AMP-responsive element-binding protein 3-like protein 3 (CREB-H).

Application

Quality Control Testing

Evaluated by Western Blotting in lysate from wild-type Mouse liver tissue.

Western Blotting Analysis (WB): A 1:1,000 dilution of this antibody detected CREB-H in lysate from wild-type Mouse liver tissue, but not in lysate from Mouse liver with CREB-H knockout.

Tested Applications

Description de la cible

Cyclic AMP-responsive element-binding protein 3-like protein 3 (UniProt: Q68CJ9; also known as cAMP-responsive element-binding protein 3-like protein 3, Transcription factor CREB-H) is encoded by the CREB3L3 (also known as CREBH, HYST1481) gene (Gene ID: 84699) in human. CREB-H is a single-pass type II membrane protein that is exclusively expressed in the liver. It contains a cytoplasmic domain (aa 1-322), a transmembrane domain (aa 323-343), and a lumenal domain (aa 344-461). It is a RIP-regulated liver-specific transcription factor that is cleaved upon endoplasmic reticulum (ER) stress and is required to activate expression of acute phase response genes. In response to ER stress, CREB-H is cleaved by site-1 and site-2 proteases to liberate an N-terminal fragment that transits to the nucleus to activate transcription of the genes encoding serum amyloid P-component and C-reactive protein. Proinflammatory cytokines are also shown to increase the expression of ER membrane-anchored CREB-H. It is activated in response to cAMP stimulation and is reported to bind to the cAMP response element (CRE) and box-B element. It plays a crucial role in the regulation of triglyceride metabolism and is required for the maintenance of normal plasma triglyceride levels. CREB-H is shown to interact with PPAR to regulate FGF21, a hepatic hormone that regulates whole-body energy homeostasis. CREB-H and PPAR are reported to act as interactive trans-activators that regulate each other for their expression. They form a functional complex upon fasting or the atherogenic high-fat diet. CREB-H-deficient mice display hypertriglyceridemia secondary to inefficient triglyceride clearance catalyzed by lipoprotein lipase. (Ref.: Kim, H., et al. (2014). Forme physique

Purified rabbit polyclonal antibody in PBS with 50% glycerol and without azide.

Reconstitution

1.0 mg/mL. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Stockage et stabilité

Store at -10°C to -25°C. Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
