General description

Decarboxylase broth base, Moeller is a dehydrated culture medium designedfor the differentiation of Gram-negative enteric bacteria based on theirability to decarboxylate specific amino acids. The medium follows the Moellerformulation, which contains peptone, beef extract, glucose, pyridoxal (as acofactor), and bromocresol purple as a pH indicator, but no amino acids bydefault; appropriate L‑amino acids (L‑arginine, L‑lysine, or L‑ornithine) are added separately depending on the test required. Wheninoculated and overlaid with mineral oil to ensure anaerobic conditions,decarboxylase-positive organisms produce alkaline amines that raise the pH andshift the indicator from yellow (acidic) back to purple (alkaline), allowingdistinction between decarboxylase-positive and -negative strains.

Application

Decarboxylase broth base, Moeller is used primarily in clinical and industrial microbiology laboratories for biochemical identification of Enterobacteriaceae and other Gram‑negative rods through detection of lysine, ornithine, or arginine decarboxylase activity. It is particularly valuable in differentiating closely related taxa such as Klebsiella and Enterobacter species (via ornithine decarboxylase) and in supporting standard identification schemes for Salmonella, Shigella, and other non-fermentative Gram‑negative bacteria. The test can be integrated into wider biochemical panels for organism profiling in clinical diagnostics, food microbiology, and environmental testing.

Preparation Note

Suspend 10.52 g of Decarboxylase Broth Base in 1000 ml of distilled water. Add 10 g of either L-Arginine (Cat. No. 11010), or L-Lysine (Cat. No. 62840), or L-Ornithine (Cat. No. 75470). Heat to dissolve the medium completely. When L-ornithine is added, readjust the pH. Dispense in 5 ml quantities in screwcapped tubes. Sterilize by autoclaving at 15 lbs. pressure (121°C) for 10 minutes.
