Undiluted serum.

Reconstitution

Reconstitute the lyophilized antibody with 100 µl of sterile distilled H₂O. Reconstitute the entire contents of the vial; during shipment and handling portions of the lyophilized pellet may have become dislodged and may not be in the bottom of the vial. Following reconstitution, aliquot and freeze (-20°C). Stock solutions are stable for up to 6 months at -20°C. Further dilute with sterile PBS or Tris buffer to dilutions no higher than 1:10.

Antibody specificity was examined in the rat cortex, hypothalamus, raphe nuclei, and spinal cord. Staining is completely eliminated by pretreatment of antibody with Serotonin (5-HT) Transporter Control Peptide (Cat. No. PP87) at a concentration of 5 µg/ml. Low levels of glutaraldehyde (0.1%-0.3%) may be used in conjunction with paraformaldehyde for tissue perfusion fixation. The specificity of this antibody was also determined by immunoblotting analysis using rat brain extracts of the cortex, hypothalamus, midbrain, and hindbrain, which specifically detected a single band. Antibody should be titrated for optimal results in individual systems.
