Spécificité

Clone 1D10 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects CD33/Siglec-3. It targets an epitope within 16 amino acids from the extracellular domain.

Application

Quality Control Testing

Evaluated by Western Blotting in U937 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected CD33/Siglec-3 in U937 cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected CD33/Siglec-3 in lysates from EL4 cells and Rat spleen tissue, and His-tagged recombinant fragment of Human CD33/Siglec-3.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected CD33/Siglec-3 in Human tonsil tissue sections.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected CD33/Siglec-3 in THP-1 cells.

Flow Cytometry Analysis: 1 µg from a representative lot detected CD33/Siglec-3 in one million Human peripheral blood mononuclear cells (PBMC).

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Description de la cible

Myeloid cell surface antigen CD33 (UniProt: P20138; also known as Sialic acid-binding Ig-like lectin 3, Siglec-3, gp67, CD33) is encoded by the CD33 (also known as SIGLEC3) gene (Gene ID: 945) in human. CD33 is a disulfide-linked homodimeric, single-pass type I membrane glycoprotein that is synthesized with a signal peptide (aa 1-17), which is subsequently cleaved off to generate the mature form that contains an extracellular domain (aa 18-259), a transmembrane domain (aa 260-282), and a cytoplasmic domain (aa 283-364). In its extracellular region it contains one Ig-like V-type domain (aa 19-135) and an Ig-Like C2 type domain (aa 145-228). In its cytoplasmic domain it has two ITIM motifs (aa 338-343 and 356-361) that are involved in modulation of cellular responses. These motifs can be phosphorylated by Src-like kinases and phosphorylated ITIM motifs can bind to SH2 domains of several SH2-containing phosphatases. Phosphorylation of CD33 by LCK on tyrosine 340 is involved in its binding to PTPN6 and PTPN11. CD33 is mainly expressed in monocytic/myeloid lineage cells. Its expression is also observed on microglial cells in the brain. It plays a role in mediating cell-cell interactions and in maintaining immune cells in a resting state. It is present in a vast majority of subjects with Acute Myeloid Leukemia (AML) but is absent from pluripotent hematopoietic stem cells. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Forme physique

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Stockage et stabilité

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
