Spécificité

Clone 5C18 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects NMDAR2B. It targets an epitope within 20 amino acids from the C-terminal, cytoplasmic domain.

Application

Quality Control Testing

Evaluated by Western Blotting in Mouse Brain tissue lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected NMDAR2B in Mouse Brain tissue lysate.

Tested applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected NMDAR2B in Rat Brain tissue lysate.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected NMDAR2B in rat brain tissue sections.

Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected NMDAR2B in E18 rat cortex primary cell cultures.

Affinity Binding Assay: A representative lot of this antibody bound NMDAR2B with a KD of 7.4 x 10-9 in an affinity binding assay.

Description de la cible

Glutamate receptor ionotropic, NMDA 2B (UniProt: Q01097; also known as GluN2B, Glutamate [NMDA] receptor subunit epsilon-2, N-methyl D-aspartate receptor subtype 2B, NMDAR2B, NR2B) is encoded by the Grin2b gene (Gene ID: 14812) in murine species. NMDAR2B It is a component of NMDA receptor complexes that function as heterotetrameric, ligand-gated ion channels with high calcium permeability and voltage-dependent sensitivity to magnesium. It is involved in long-term potentiation and neuronal plasticity and regulate the number of nerve cells during development; however, over-stimulation of NMDA receptors can lead to neuronal degeneration. NMDAR2B is a heterotetrameric, multi-pass membrane protein that is synthesized with a signal peptide (aa 1-26), which is subsequently cleaved off in the mature form that contains two extracellular domains (aa 27-557 and 647-817), three transmembrane domains, and three cytoplasmic domains. It contains two zeta subunits and two epsilon subunits but can also form channels that contain at least one zeta subunit and at least one epsilon subunit. Activation of NMDAR2B channel requires binding of glutamate to the epsilon subunit, glycine binding to the zeta subunit, plus membrane depolarization to eliminate channel inhibition by magnesium ions. NMDAR2B is reported to be phosphorylated at Ser 1303 by DAPK1 that enhances its channel activity allowing injurious Ca2+ influx that results in neuronal death. Mutations in NMDAR2B in humans have been linked to early infantile epileptic encephalopathy that is characterized by refractory seizures and neurodevelopmental impairment. Some mutations are also known to cause mental retardation with or without seizures. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Forme physique

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Stockage et stabilité

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
