Informations sur le gène

human ... KLHL20(27252)

Description générale

The gene KLHL20 (kelch like family member 20) is mapped to human chromosome 1q25.1. The encoded protein localizes to the trans-Golgi network (TGN). Small amount of the protein is also present in PML-NBs (promyelocytic leukemia - nuclear bodies). KLHL20 belongs to the BTB (broad-complex, tramtrack, and bric-à-brac) family of proteins.

Immunogène

Recombinant fragment contain a sequence corresponding to a region within amino acids 1 and 609 of KLHL20 according to NP_055273

Actions biochimiques/physiologiques

KLHL20 (kelch like family member 20) is a substrate adaptor of cullin3 (Cul3). It is required for post-Golgi trafficking. The Cul3-KLHL20 complex works as an ubiquitin E3 ligase and is responsible for polyubiquitination of coronin-7, thereby targeting coronin-7 to trans-Golgi network. KLHL20 is also involved in autophagy by regulating autophagy-associated degradation of ULK1 (unc-51 like autophagy activating kinase 1) and VPS34 (vacuolar protein sorting 34) complex subunits. Additionally, KLHL20 is associated with hypoxia responses and tumorigenesis. HIF1 (hypoxia inducible factor 1)-mediated upregulation of KLHL20 gene causes hypoxia-induced PML (promyelocytic leukemia) polyubiquitination. PML destruction is linked with epithelial-mesenchymal transition, tumor growth, angiogenesis and other related events.

Knowledge about the genetic lesions that occur in Burkitt's lymphoma, besides the pathognomonic IG-MYC translocations, is limited. Thirty-nine molecularly-defined Burkitt's lymphomas were analyzed with high-resolution single-nucleotide polymorphism chips for genomic imbalances and uniparental disomy. Imbalances were correlated to expression profiles

Ubiquitin chains are formed as structurally distinct polymers via different linkages, and several chain types including K33-linkage remain uncharacterized. Here, we describe a role for K33-polyubiquitination in protein trafficking. We show that the Cullin 3 (Cul3) substrate adaptor KLHL20 is
