General description

Fontana-Masson staining is used for the demonstration of argentaffin substances such as melanin, argentaffin granules and some neurosecretory granules.

Biochem/physiol Actions

Argentaffin cells and melanin possess the ability to bind silver from a silver solution and reduce to visible metallic silver without the need for a reducing agent. The Fontana-Masson Stain Kit can be used for frozen or paraffin embedded sections.

Questions

1–2 of 2 Questions
  1. Is there a procedure or information available for the use of the Fontana Masson staining kit in 3D tissue structures for the quantification of Neuromelanin?

    1 answer
    1. The HT200 kit is approved for IVD use and is specifically approved for use with standard paraffin-embedded tissue cut at 4 or 5 microns. No procedures have been developed for possible use in 3D tissue structures for the quantification of Neuromelanin. It would be required to adapt the procedure for any use not outlined in the Instructions for Use. The time spent in the ammoniacal silver stain solution would need to be adjusted, and it is suggested to start at 15 minutes and proceed in increments of 15 minutes until acceptable staining is obtained for staining 3D tissue. If results are promising, it is recommended to possibly proceed in increments of 5 minutes to optimize the staining results. It is possible that the staining times for the other steps may also need to be adjusted.

  2. Is it possible to collect viable DNA from tissue stained with the Fontanna Masson HT200 kit?

    1 answer
    1. Collecting DNA and RNA from FFPE fixed tissues is a common practice, typically involving tissue from paraffin-embedded blocks or slides stained with Hematoxylin and Eosin. Collecting DNA and RNA from FFPE fixed tissues is a common practice, typically involving tissue from paraffin-embedded blocks or slides stained with Hematoxylin and Eosin.
