General description
L′enzyme de conversion de l'angiotensine (ECA) est une dipeptidyl-carboxypeptidase possédant deux isoformes (somatique et testiculaire) contenant un motif de liaison au zinc du type HEXXH dans leur site actif. L′ECA régule la pression artérielle via le système rénine-angiotensine. Elle augmente la pression artérielle en convertissant l′angiotensine I en angiotensine II, un vasoconstricteur essentiel, et en inhibiant la bradykinine, un vasodilatateur puissant. L′inhibition de l′ECA constitue une stratégie thérapeutique en cas de forte pression artérielle. Il existe plusieurs peptides inhibiteurs synthétiques de l′ECA pour un usage clinique, notamment le captopril, l′énalapril et le lisinopril. Des peptides inhibiteurs destinés à inhiber l′ECA sont actuellement développés à partir d′aliments naturels ou de composés phénoliques d′origine végétale. L′ECA joue un rôle déterminant dans la fécondation, en libérant les protéines ancrées dans le glycosylphosphatidylinositol (GPI) de la membrane du sperme.
Application
L′enzyme de conversion de l′angiotensine issue de poumon de lapin a été utilisée :
- Pour mesurer l′effet inhibiteur d′hydrolysats de protéines de blanc d′oeuf sur l′activité de l′ECA par chromatographie liquide de haute performance (HPLC).
- Pour mesurer l′inhibition de l′ECA par du péricarpe de litchi et du blanc de poulet cuit sur un substrat d′hippuryl-L-histidyl-L-leucine (HHL) par HPLC en phase inverse (RP-HPLC) et HPLC, respectivement.
- Pour libérer les protéines ancrées dans le GPI in vitro dans quelques lignées cellulaires telles que HeLa et HEK293, et in vivo dans du sperme de souris.

Élimine les dipeptides C-terminaux des substrats cibles, par exemple l′angiotensine I et la bradykinine.
