Description générale

Purified mouse monoclonal antibody derived by immunizing BALB/c mice with the specified immunogen and fusing splenocytes with SP2/0 mouse myeloma cells. Recognizes N-myc as well as its cleavage products.
Recognizes N-myc as well as its cleavage products in IMR5 cells. Myc is a labile protein. Fresh lysates in the presence of protease inhibitor cocktails are recommended.
The v-myc oncogene, initially identified in the MC29 avian retrovirus, causes myelocytomas, carcinomas, sarcomas and lymphomas, and belongs to a family of oncogenes conserved throughout evolution. In humans the family consists of five genes: c-myc, N-myc, R-myc, L-myc and B-myc. Amplification of the N-myc gene has been found in human neuroblastomas and cell lines. The extent of N-myc amplification correlates well with the stage of neuroblastoma disease. Immunological studies have shown that the human N-myc gene gives rise to at least two nuclear phosphoproteins of ~64 kDa and ~67 kDa that exhibit relatively short (30 min) half lives in vivo and exhibit DNA binding properties in vitro.

Immunogène

recombinant, human, N-Myc fusion protein, expressed in E. coli

Application

Frozen Sections (1-5 μg/ml)
Immunoblotting (1-5 μg/ml, see application references)
Immunofluorescence (1-5 μg/ml, see application references)
Immunoprecipitation (1-2 μg/sample, see application references)

Forme physique

Lyophilized from a volatile buffer, 100 μg BSA.

Reconstitution

Reconstitute the lyophilized antibody with 1 ml sterile PBS. Lyophilized antibodies should be reconstituted at 4°C with occasional gentle mixing for at least 2 h. Following reconstitution, refrigerate (4°C) for short-term storage or aliquot and freeze (-2

Remarque sur l'analyse

Positive Control
IMR5 cells

Autres remarques

It is important to compensate for the short half life of this protein when preparing tissue sections or extracts. We recommended that all preparations be kept cold and that a cocktail of protease inhibitors be used. Antibody should be titrated for optimal

Code de la classe de stockage

The human N-myc gene is related to the c-myc proto-oncogene, and has been shown to have transforming potential in vitro. Many studies have reported amplification of N-myc in human neuroblastoma and retinoblastoma cell lines. In primary tumors, amplification of the

A domain of DNA designated N-myc is amplified 20- to 140-fold in human neuroblastoma cell lines but not in cell lines from other tumor types. N-myc has now been found to be amplified in neuroblastoma tissue from 24 of 63
