Specificity

Clone 1D5 is a ZooMAb® rabbit recombinant monoclonal antibody that detects PABP-1. It targets an epitope within 21 amino acids from the C-terminal part.

Immunogen

KLH-conjugated linear peptide corresponding to 21 amino acids from the C-terminal region of human PABP-1.

Application

Quality Control Testing

Evaluated by Western Blotting in A549 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected PABP-1 in A549 cell lysate.

Tested Applications

Western Blotting Analysis: A representative lot of this antibody detected PABP-1 in A549, NIH/3T3 cell lysates (1:1,000 dilution), and human testis lysate (1:10,000 dilution).

Flow Cytometry Analysis: 1 µg from a representative lot detected PABP-1 in one million HEK-293 cells .
Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected PABP-1 in human testis tissue sections.

Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected PABP-1 in HEK-293 cells.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Polyadenylate-binding protein 1 (UniProt: P11940; also known as PABP-1; Poly(A)-binding protein 1) is encoded by the PABPC1 (also known as PAB1;PABP;PABP1, PABPC2) gene (Gene ID: 26986) in human. Polyadenylate-binding protein 1 (PABP1) is a multifunctional protein characterized by four RNA recognition motifs (RRMs) that bind to the poly(A) tail of mRNA molecules. Additionally, PABP1 has a proline-rich C-terminal domain involved in protein-protein interactions, which is essential for its role in regulating mRNA metabolic processes. PABP1 plays a critical role in the regulation of mRNA stability and translation. By binding to the poly(A) tail of mRNAs, it enhances the stability of the mRNA and facilitates its efficient translation by ribosomes. PABP1 is also involved in the modulation of mRNA polyadenylation during transcription and influences mRNA nuclear export and deadenylation. PABP1 is ubiquitously expressed across various tissues, reflecting its fundamental role in mRNA metabolism. The expression levels of PABP1 can vary depending on cellular conditions and stress, indicating its involvement in adaptive cellular responses. Dysfunction or abnormal levels of PABP1 are associated with several human diseases. For instance, altered PABP1 expression is linked to cancer, where it can affect cell proliferation and tumor progression by modulating the stability and translation of mRNAs encoding oncogenes and tumor suppressors. Additionally, PABP1 has been implicated in viral infections; many viruses manipulate PABP1 to preferentially translate viral mRNAs. Neurodegenerative diseases such as Amyotrophic Lateral Sclerosis and Frontotemporal Lobar Degeneration have also been associated with mutations in the PABP1 gene, affecting its normal function.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
