Specificity

Clone 1I13 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects JAK1. It targets an epitope within 21 amino acids from the N-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 21 amino acids from the N-terminal half of human Janus kinase 1 (JAK1).

Application

Quality Control Testing

Evaluated by Western Blotting in HeLa cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected JAK1 in HeLa cell lysate.

Tested applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected JAK1 in C6, COS-7, and Ramos cell lysate.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected JAK1 in human kidney tissue sections.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected JAK1 in Ramos cells.

Affinity Binding Assay: A representative lot of this antibody bound JAK1 peptide with a KD of 1.9 x 10-7 in an affinity binding assay.

Target description

Tyrosine-protein kinase JAK1 (UniProt: EC:2.7.10.2, Janus kinase 1, JAK-1) is encoded by the JAK1 (also known as JAK1A, JAK1B) gene (Gene ID: 3716) in human. The Janus kinase (JAK)/signal transducers and activators of transcription (STAT) signaling pathway play an important role in cell proliferation, cell differentiation, cell migration, and cell death. It is the principal signaling mechanism for a variety of cytokines and growth factors. In mammals, the JAK family is composed of four members: JAK1, JAK2, JAK 3, and Tyk2. In their activation scheme, upon ligand binding, the two JAKs into close proximity, allowing their transphosphorylation and the activated JAKs can then phosphorylate STAT proteins. JAK1 is a non-receptor, cytosolic tyrosine kinase with a multi-domain structure that includes a C-terminal tyrosine kinase domain (aa 873-1153) that is flanked by a catalytically inactive pseudokinase domain (aa 583-855) with regulatory function. Their N-terminal half contains a FERM homology domain (aa 34-420) that is implicated in receptor binding, and an adjacent SH2 domain (aa 439-544). The FERM and SH2 domains constitute the receptor-binding module and the pseudokinase domain is thought to regulate kinase activity. Somatic mutations in JAK1 are reported in cases of acute lymphoblastic leukemia (ALL) and are associated with poor response to therapy and overall prognosis. In mice, complete lack of JAK1 leads to perinatal lethality and reduction in thymocytes and B cells. This ZooMAbZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
