Specificity

Clone 1H18 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Early endosome antigen 1 (EEA1). It targets an epitope within 24 amino acids from the N-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 24 amino acids from the N-terminal half of human Early endosome antigen 1 (EEA1).

Application

Quality Control Testing

Evaluated by Western Blotting in HeLa cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected Early endosome antigen 1 (EEA1) in HeLa cell lysate.

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected Early endosome antigen 1 (EEA1) in PC12, SHSY5Y, and HepG2 cell lysates.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected Early endosome antigen 1 (EEA1) in A431, HeLa and HepG2 cells.s.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected Early endosome antigen 1 (EEA1) in human cerebral cortex tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound Early endosome antigen 1 (EEA1) with a KD of 1 x 10-12 in an affinity binding assay.

Anti-EEA1, clone 1H18 ZooMAb®, Cat. No. ZRB1592, is a recombinant Rabbit monoclonal antibody that detects Early endosome antigen 1 (EEA1) and used in Affinity Binding Assay, Immunocytochemistry, Immunohistochemistry (Paraffin), and Western Blotting.

Target description

Early endosome antigen 1 (UniProt: Q15075; also known as Endosome-associated protein p162, Zinc finger FYVE domain-containing protein 2, EEA1) is encoded by the EEA1 (also known as ZFYVE2) gene (Gene ID: 8411) in human. EEA1 is a homodimeric peripheral membrane protein that binds to phospholipid vesicles containing phosphatidylinositol 3-phosphate and participates in endosomal trafficking. It binds to Rab5 proteins that have been activated by GTP-binding. The stable association of EEA1 with the endosomal membrane is regulated by PI3 kinase, Rab5 and Ca2+/calmodulin. At its carboxy terminal, it contains a PYVE type zinc finger domain (aa 1352-1410) that mediates its interaction with phosphatidylinositol 3-phosphate in membranes of early endosomes and penetrates bilayers. The FYVE domain insertion into phosphatidylinositol 3-phosphate-enriched membranes is shown to substantially increase in acidic conditions. When endosome fusion is not required, EEA1 serves as a substrate for p97, promoting EEA1 dissociation and endosome independence. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb® formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
