Biochem/physiol Actions

TAT-SAMβA is a mitofusin 1 (MFN1)-βIIPKC association blocking peptide that prevents βIIPKC-mediated negative regulation of MFN1. TAT-SAMβA protects neonatal and adult rat cardiac myocytes, but not Mfn1-knockout cells, from stress-induced death in cultures and improves cardiac contractility in rats with heart failure in vivo (3 mg/kg/day for 6 wks via Alzet pump delivery) by re-establishing mitochondrial morphology and function. TAT-SAMβA is composed of a 9-aa βIIPKC-derived MFN1-interacting sequence linked at the N-terminal to the cell permeable TAT(47-57) via a -Gly-Gly- spacer
