General description

CNOT3 (CCR4-NOT transcription complex subunit 3) is a member of the conserved multi-protein complex called CCR4-NOT. This gene is localized to human chromosome 19q13.4. It contains the conserved Not-Box motif in its C-terminal.

Application

Biochem/physiol Actions

CNOT3 (CCR4-NOT transcription complex subunit 3) is a part of CCR4-Not complex, which plays an essential role in the control of gene expression. It negatively regulates gene transcription by either directly interacting with the promoter of the target gene, or by modulating the rate of target mRNA degradation. It directly interacts with and suppresses PRPF31 (Pre-MRNA Processing Factor 31) expression. Thus, it modulates PRPF31 mutations in retinitis pigmentosa. It also functions as a tumor suppressor, and is mutated in certain cases of adult T-cell acute lymphoblastic leukemia (T-ALL). It plays an essential role in maintaining the embryonic stem cell (ESC) properties in humans and mice. It prevents ES differentiation into extra-embryonic lineages.
