Cell Line Description
The cell line Xrs6-hamKu80 was derived from a radiosensitive CHO-K1 mutant transfected with hamster Ku80 cDNA. Ku80, a subunit of the Ku protein and component of the DNA-dependent protein kinase, is a gene product defective in radiosensitive CHO cells, so called xrs mutants. XRC5, the gene encoding Ku80, belongs to the ionisation radiation group affected in DNA repair defects. Mutants transfected with hamster Ku80 obtained full restoration of gamma-ray resistance, DNA end-binding activity to parent levels and expression of Ku80 and Ku70 proteins.
Split sub-confluent cultures (70-80%) 1:3 to 1:6 i.e. seeding at 2-4x10,000 cells/cm2 using 0.25% trypsin or trypsin/EDTA; 5% CO2; 37°C.
