Cell Line Origin

Mouse alveolar macrophage, SV40 transformed

Cell Line Description

The MH-S cell line was obtained from a 7 week-old male BALB/c J mouse by bronchoalveolar lavage followed by SV-40 transformation of an adherent cell enriched population of alveolar macrophages. Many properties of alveolar macrophages are expressed with the cells being phagocytic, esterase positive and peroxidase negative. The intracellular T-antigen and cell surface Ia and Mac-1 antigens are expressed as well as Fc receptors. IL-1 is constitutively expressed and can be increased by stimulation with lipopolysaccharides (LPS). A cellulose dependant suppression of the in vitro plaque-forming cell (PFC) i-espouse by MH-S cells has also been reported.

Application

Phagocytosis studies, plaque assays, IL-1 assays

Culture Medium

RPMI 1640 + 2mM Glutamine + 0.05mM 2-Mercaptoethanol (2ME) + 10% Foetal Bovine Serum (FBS).

Subculture Routine

Split sub-confluent cultures (70-80%) 1:2 to 1:4 i.e. seeding at 1-2x10,000 cells/cm2 using 0.25% trypsin or trypsin/EDTA; 5% CO2; 37°C.
