General description

Chemical structure: peptide

Application

Colistin sulfate is used to permeabilize bacterial cell membranes and to study mannose-resistant haemagglutination and antibiotic resistance in certain organisms such as A. baumannii [1]. It has been used to study hephrotoxicity in the rat kidney [2], and MICs, time-kill kinetics, and postantibiotic effect (PAE) against Pseudomonas aeruginosa [3].

Biochem/physiol Actions

Mode of Action: Binds to lipids on the cell cytoplasmic membrane of Gram-negative bacteria and disrupts the cell wall integrity.
Antimicrobial spectrum: Gram-negative bacteria.
Mode of Action: Binds to lipids on the cell cytoplasmic membrane of Gram-negative bacteria and disrupts the cell wall integrity.
Antimicrobial spectrum: Gram-negative bacteria. It is proposed that renal reabsorption of colistin may involve organic cation transporters and peptide transporters and that the process is sensitive to pH [2].

Analysis Note

Freely soluble in water, practically insoluble in acetone and in ethanol (96%)

Other Notes

Keep container tightly closed in a dry and well-ventilated place.

Acinetobacter baumannii is a prevalent nosocomial pathogen with a high incidence of multidrug resistance. Treatment of infections due to this organism with colistin, a last-resort antibiotic of the polymyxin class, can result in the emergence of colistin-resistant strains. Colistin resistance

Infections caused by multidrug-resistant (MDR) bacteria, particularly Gram-negative bacteria, are an escalating global health threat. Often clinicians are forced to administer the last-resort antibiotic colistin; however, colistin resistance is becoming increasingly prevalent, giving rise to the potential for a situation

Drug resistance is a public health concern that threatens to undermine decades of medical progress. ESKAPE pathogens cause most nosocomial infections, and are frequently resistant to carbapenem antibiotics, usually leaving tigecycline and colistin as the last treatment options. However, increasing

Polymyxin-based combination therapy is often used to treat carbapenem-resistant Acinetobacter baumannii (A. baumannii) infections. Although sulbactam is intrinsically active against A. baumannii, few studies have investigated colistin/sulbactam combinations against carbapenem-resistant A. baumannii. Whole genome sequencing was undertaken on eight carbapenem-resistant (colistin-susceptible)

Persisters are small populations of quiescent bacterial cells that survive exposure to bactericidal antibiotics and are responsible for many persistent infections and posttreatment relapses. However, little is known about how to effectively kill persister bacteria. In the work presented here

Questions

1–8 of 8 Questions
  1. 1. What is the solubility of this antibiotic in water? How many mg of the substance can dissolve in 1 mL of water? 2. After preparing a diluted solution of the antibiotic in water, where should it be stored, and what is the recommended storage duration?

    1 answer
    1. This material is quite soluble in water. The rate of solubility in water has not been established. However, other sources report rates up to 560 mg/mL. Solutions prepared in water are stable when refrigerated at 2-8 °C for at least 60 days. Avoid storing solutions prepared in isotonic buffers, as the literature indicates a possible increase in the rate of decomposition.

  2. How is shipping temperature determined? And how is it related to the product storage temperature?

    1 answer
    1. Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature. As shipping routes are configured for minimum transit times, shipping at ambient temperature helps control shipping costs for our customers. 3. How can I determine the shelf life / expiration / retest date of this product?

    1 answer
    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.

  4. what is potency in microgram/mg?

    1 answer
    1. As per the International Standard Preparation (1968), for comparative purposes, each microgram (ug) of colistin base is equivalent to 30 units. As an example, a lot reporting an activity of 22,891 units/mg would have the equivalent of 763 ug of colistin base per mg solid. This product has a minimum activity specification of 19,000 IU/mg, which equates to 633 ug colistin base per mg solid. The activity is reported on a lot specific basis on the product Certificate of Analysis. Please see the link below to review a sample or lot specific Certificate.
