Specificity

Clone C8Mab-2 is a rat monoclonal antibody that detects C-C chemokine receptor type 8 (CCR8). It targets an epitope within the extracellular domain.

Immunogen

Chinese Hamster Ovary (CHO) cells stably expressing mouse CCR8.

Application

Quality Control Testing

Evaluated by Flow Cytometry in Chinese Hamster ovary (CHO) cells stably expressing murine CCR8.

Flow Cytometry Analysis: 1.0 µg of this antibody detected CCR8 in one million Chinese Hamster ovary CHO) cells stably expressing murine CCR8.

Target description

C-C chemokine receptor type 8 (UniProt: P56484; also known as C-C CKR-8; CC-CKR-8; CCR-8, CDw198) is encoded by the Ccr8 (also known as Cmkbr8, Ter1) gene (Gene ID: 12776) in murine species. CCR8, a member of the G-protein coupled receptor 1 family, is a multi-pass membrane protein with four extracellular domains, seven transmembrane domains, and four cytoplasmic domains. Its expression is observed in thymus and its higher expression is observed in Treg and T helper 2 (TH2) cells. It is recruited to the inflammation site and has been implicated in allergy and asthma. It serves as a receptor for CCL1/SCY1 chemokine. In human and in murine models CCR8+ Tregs account for 30 to 80% of tumor Tregs in various cancers and only less than 10% in other tissues. However, most tumor-infiltrating conventional T cells are reported to be CCR8 negative. Disruption of CCR8 function is reported to result in reduced accumulation of Treg cells within tumors and disrupts their immunosuppressive function. (Ref.: Kidani, Y., et al. (2022). Proc. Natl.
