General description
cGMP-dependent 3′,5′-cyclic phosphodiesterase (UniProt: O00408; also known as EC:3.1.4.17, PDE2A, Cyclic GMP-stimulated phosphodiesterase, CGS-PDE, cGSPDE) is encoded by the PDE2A gene ( Gene ID: 5138) in human. PDE2A is an evolutionarily conserved cyclic nucleotide phosphodiesterase with a dual-specificity for the second messengers cAMP and cGMP that are key regulators of many important physiological processes. PDE2A is expressed in both the periphery and in the central nervous system, however its expression is highest in the brain, where it is mainly localized in the cortex, hippocampus, and striatum. Three splice variants of PDE2A have been described, the PDE2A1, PDE2A2 and PDE2A3. PDE2A1 is cytosolic whereas -A2 and -A3 are membrane bound. PDE2A isoform 2 is specifically targeted to the mitochondrial matrix, where it forms a local signaling system with soluble adenylate cyclase regulating the respiratory chain. PDE2A is a dimeric enzyme that binds 2 divalent metal cations per subunit. Site 1 may preferentially bind zinc ions, while site 2 has a preference for magnesium and/or manganese ions. Each PDE2A monomer contains an N-terminal (200 residues) domain of unknown function, tandem GAF domains (GAF A and GAF B), and a C-terminal catalytic domain. GAF A functions as a dimerization locus. PDE2 A catalytic activity is allosterically stimulated by cGMP binding to its GAF B domain. Binding of cGMP induces conformational change in the protein structure leading to higher enzyme activity.
