General description

Human recombinant histone H3.3-H4 tetramers expressed in E Coli. as fusion proteins with either an N-terminal FLAG® or 6xHis epitopes, and purified by Fast Protein Liquid Chromatography (FPLC).

The core histones, H2A, H2B, H3, and H4, form a central octameric structure that binds approximately 147 bp of DNA to form a nucleosome. The histone variant H3.3 replaces the conventional H3.1 histone in a wide range of nucleosomes in active genes, and are incorporated into chromatin independently of DNA synthesis. The structure of chromatin can be altered by post-translational modifications (PTM) occurring on the C- and N-terminal tails of core histones. Histone modifications can either provide or hinder access of key transcriptional complexes to DNA sequences and can regulate the expression of genes. These effects can be deciphered by applications such as chromatin immunoprecipiation (ChIP) which requires high quality chromatin and essential tools such as histone proteins and appropriate enzymes to assemble chromatin in vitro.
