General description

Interleukin-18 (UniProt: Q14116; also known as IL-18, Iboctadekin, Interferon gamma-inducing factor, IFN-gamma-inducing factor, Interleukin-1 gamma, IL-1 gamma) is encoded by the IL18 (also known as IGIF, IL1F4) gene (Gene ID: 3606) in human. IL-18 is a pro-inflammatory cytokine that is shown to evoke both innate and acquired responses. It is synthesized with a propeptide (aa 1-36), which is subsequently cleaved off by caspases 1 and 4 to generate the active mature form. IL-18 is reported to be constitutively expressed in monocytes, macrophages, epithelial cells, osteoblasts, keratinocytes, and synovial fibroblasts. Its expression has also been reported in adrenal cortical cells. Mature IL-18 acts by binding to the IL-18 receptor α (IL-18α) to form a low affinity binary complex that recruits IL-18β to form a high affinity complex that leads to the activation of NF-kB and MAP kinase pathways. IL-18 can synergize with IL-12 to induce interferon-γ synthesis from T-helper 1 and natural killer cells.
