General description

Hemoglobin is the major component of red blood cells, and is responsible for their red color. Its normal concentration in erythrocytes is 34%. Hemoglobin is the most important respiratory protein of vertebrates by virtue of its ability to transport oxygen from the lungs to body tissues, and to facilitate the return transport of carbon dioxide.

Application

Hemoglobin was used in the development of a rapid enzyme immunoassay for the detection of retinol-binding protein.[1] It was also used in the inhibition of human platelet reactivity by endothelium-derived relaxing factor.[2]

Biochem/physiol Actions

The Fe2+/Fe3+ balance is a physiological indicator of blood oxygenation. Deoxygenated hemoglobin accessorizes a feedback loop by reducing nitrite to NO, a vasodilator which enhances blood flow to oxygen-deprived tissues.

Caution

Since native hemoglobin is readily oxidized in air, these preparations may be predominantly methemoglobin.

    1. This product is a crude preparation. The purity has not been determined. Unfortunately, purified human hemoglobin is not available at this time.

  3. Is it LPS-free? when i use this hemoglobin for cell treatment, should I treat polymyxin b B also?

    1. This product is not tested for endotoxin. For use with applications that may be sensitive to presence of LPS, pre-treatment with polymyxin B is a viable approach and is known to effectively neutralize endotoxins.   4. What are the reconstitution instructions for the product, including the amount of liquid and suggested concentration? Additionally, what is the stability of the product after reconstitution?

    1. The product H7379 is tested for solubility at 20 mg/ml during quality control testing. In the past, there was a hemoglobin standard available at 18g/dL or 18% in lyophilized form, mixed with Drabkins reagent to produce a stable cyanomethemoglobin solution. This reagent was stated to be stable for 6 months, with notes indicating that lower concentrations were not necessarily stable. The absorbance values for this solution are typically read at 540 nm.

      After reconstitution, if blood hemoglobin levels are not being determined, it is suggested to take the absorbance in water, scanning the wavelengths from 500-700 nm. A dilution should be performed to achieve an absorbance within the general range of 0.7 to 1.5. After measuring the absorbance of the freshly prepared solution, a baseline is established, and periodic measurements of the absorbance can be taken to determine if the values are trending lower, indicating the need to prepare a fresh solution.

  5. What is the hemoglobin concentration in the product H7379-1G?

    1. The Specification Sheet for H7379 does not list any purity or concentration. The product is assumed to be 100% hemoglobin. Therefore, if 1 mg of H7379 is dissolved in 1 ml of deionized water, the concentration would be 1 mg/ml.

  7. I bought this product in June 2022 and have been using it and storing it properly and was wondering the shelf life of this Product H739-5G

    1. This product does not contain an expiration date, it contains an recommended retest date of roughly 4 years. A recommended retest date is the period of time during which the product is expected to remain within established stability specifications, provided that it has been stored under defined conditions. After the Retest Date, product samples should be examined to ensure that the product is still in compliance with the established specifications.   8. How many donors are used to make a batch of Product H7379, Hemoglobin human?

    1. This will vary with the batch size, but may come from as many as several hundred donors. All donors are tested and found negative for antibodies to HIV-1/HIV-2, HCV, for HBSAG, and for syphilis.

    1. Yes. The iron in hemoglobin will oxidize by exposure to air unless special handling and packaging is used.  Because this product is exposed to air during purification and packaging, it will be predominantly methemoglobin.
