Cell Line Origin

Human lymphoblast, HPRT - deficient

Cell Line Description

The lymphoblastoid cell line GM1899A has been derived from a Lesch-Nyhan patient. It is deficient for hypoxanthine-guanine phosphoribosyl transferase (HPRT) with a point mutation in the respective gene. This cell line is used as a feeder for extended-term culture of human T-lymphocytes (see LB lines in the collection).

Culture Medium

Dutch Modified RPMI 1640 + 2mM Glutamine + 20mM Hepes + 2mM Sodium Pyruvate (NaPy) + 20% Heated Inactivated Foetal Bovine Serum (HI-FBS)

Subculture Routine

Maintain cultures between 3-9 x 100,000 cells/ml; 5% CO2; 37°C. Do not dilute below 1 x 100,000 cells/ml, wash 2-3 times after resuscitation before seeding; disaggregate every few days by pipetting to avoid formation of large aggregates
