General description

Histone MacroH2A (mH2A) is a variant histone also known as H2AFY. mH2A is enriched in chromatin of the female inactive X chromosome and in highly methylated CpG loci. This histone variant is known to repress RNA polymerase II-driven transcription at the level of transcriptional initiation. In contrast to most histone variants containing only subtle changes, mH2A contains a large non-histone domain (the macro domain) on its C-terminus, resulting in a histone approximately three times the size of conventional H2A. There are 3 isoforms of mH2A, two of which (mH2A1.1, mH2A1.2 ) are formed by alternative splicing and another (mH2A2) from a separate gene. There are two phosphorylation sites within the basic hinge region of mH2A: Thr128 and Ser137. Phospho-Ser137 is enriched in mitosis, implicating mH2A in the regulation of posttranslational modifications throughout the cell cycle.
