General description

The BL21(DE3) pLysE Chemically Competent Cells produce T7 lysozyme to repress low-level transcription from the T7 promoter prior to IPTG induction, helping to stabilize inserts coding for deleterious products.
These cells have a higher level of repression by T7 lysozyme, offering the greatest control over expression of inserts.

  1. What is the difference between the strain expressing pLysE and pLysS?

    1. Products 69451-M and 70236-M are exactly the same competent cells. The only difference is in the packaging. The 70236 is already in 50uL aliquots, which are single-use, each aliquot is used for one reaction. This reference is available in two sizes of 11 or 22 reactions. While product 69451 is available in single 0.4 or 1mL volumes which may be thawed once and dispensed to smaller aliquots.
      The difference between the 69451-M/70236-M products and the CMC0015 BL21 is the type of plasmid that they express for the T7 lysozymes. The expression of T7 lysozyme reduces the basal expression of the target genes, inhibiting the T7 RNA polymerase. Specifically, with the expression of the plasmid pLysE (Ref. CMC0015) a higher expression of the levels of T7 Lysozime is obtained in comparison with the plasmid pLysS (ref. 69451-M/70236-M). Consequently, with reference CMC0015 there is even less risk of basal expression of the target protein in the absence of IPTG stimulation. This could be particularly relevant for toxic proteins that could affect competent cell viability.
