Gene Information

General description

DR5 (death receptor 5) or TNFRSF10B (tumor necrosis factor receptor superfamily member 10b) is a member of the tumor necrosis factor receptor (TNFR) superfamily. It is a tumor suppressor gene localized to human chromosome 8p and is composed of 10 coding exons. The encoded protein is an integral plasma membrane protein.
Recombinant human sTNFRII is an 18.9 kDa protein (174 amino acid residues) comprising the cysteine rich ligand binding portion of the extracellular domain of the TNFRII protein.

Biochem/physiol Actions

It initiates the formation of death-inducing signal complex (DISC) by recruiting fas-associated death domain (FADD) and caspases to its cytoplasmic death domain (DD). In human lung cancer cells, parthenolide (PTL) induces extrinsic apoptosis by up-regulating TNFRSF10B. Deregulation of this gene results in metastasis, invasion and migration of tumor cells. Mutation leads to loss of p arm in this gene which is frequently the case in head and neck tumors. In a breast cancer xenograft mouse model, DR5 facilitates skeletal metastasis. In prostate carcinoma (PCa) and prostatic intraepithelial neoplasia (PIN), the expression of DR5 is lower in higher grade tumors.

Physical form

Lyophilized with no additives.

Reconstitution

Centrifuge the vial prior to opening. Reconstitute in water to a concentration of 0.1-1.0 mg/ml. Do not vortex. This solution can be stored at 2-8°C for up to 1 week. For extended storage, it is recommended to further dilute in a buffer containing a carrier protein (example 0.1% BSA) and store in working aliquots at -20°C to -80°C.
