General description

Human eukaryotic translation elongation factor 1 alpha has two isoforms, alpha 1 and alpha 2. The alpha 1 isoform gene encodes a subunit of the elongation factor-1 complex which is responsible for the enzymatic delivery of aminoacyl tRNAs to the ribosome. The alpha 1 isoform is expressed in brain, placenta, lung, liver, kidney, and pancreas. The other isoform (alpha 2) is expressed in brain, heart and skeletal muscle.
The alpha 1 isoform is identified as an autoantigen in 66% of patients with Felty syndrome. The gene for the alpha 1 isoform has been found to have multiple copies on many chromosomes, some of which, if not all, represent different pseudogenes. The gene for the alpha 2 may be critical in the development of ovarian cancer.

Specificity

This antibody recognizes EF-1α (Elongation factor-1 alpha), Mr 53 kDa. Occasionally, a 38 kDa protein is also detected in A431 cell lysate.

Immunogen

Crude calmodulin-binding proteins from Trypanosoma brucei isolated using calmodulin-affinity chromatography. Clone CBP-KK1.

Application

Detect EF1α using this Anti-EF1α Antibody, clone CBP-KK1 validated for use in IP & WB.
Immunoprecipitation:
4 µg of a previous lot immunoprecipitated EF1α from an A431 RIPA cell lysate.

Quality

Routinely evaluated on RIPA lysates from non-stimulated A431 cells, human Jurkat or mouse 3T3/A31.

Western Blot Analysis:
0.5-2 µg/mL of this lot detected EF1α in RIPA lysates from non-stimulated A431 cells. Previous lots detected EF1α in human Jurkat and mouse 3T3/A31 RIPA cell lysates.

Storage and Stability

Stable for 1 year at -20°C from date of receipt.

Handling Recommendations:
Upon receipt, and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20ºC. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.

Analysis Note

Control
Positive Antigen Control: Catalog #12-301, non-stimulated A431 cell lysate. Add 2.5µL of 2-mercaptoethanol/100µL of lysate and boil for 5 minutes to reduce the preparation. Load 20µg of reduced lysate per lane for minigels.
