Clone RMST-2 is a ZooMAb® rat recombinant monoclonal antibody that specifically detects ST2/IL-33R.

Application

Quality Control Testing

Evaluated by Western Blotting in HEK293 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected ST2/IL-33R in HEK293 cell lysate.

Tested Applications

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected ST2/IL-33R in L5178Y cells transfected with ST2/IL-33R expression vector.

Flow Cytometry Analysis: 0.1 µg from a representative lot detected ST2/IL-33R in one million L5178Y cells transfected with ST2/IL-33R expression vector.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Interleukin-1 receptor-like 1 (UniProt: P14719; also known as EC:3.2.2.6, Interleukin-33 receptor alpha chain 1, Lymphocyte antigen 84, Protein ST2, Protein T1) is encoded by the Il1rl1 (also known as Ly84, St2, Ste2) gene (Gene ID: 17082) in murine species. IL-33, a cytokine that is constitutively produced from by several structural and lining cells, transmits its signal through its receptor IL-33R (ST2), which is expressed on the surface of T helper (Th2) cells and group 2 lymphoid cells. IL-33R is a single-pass type I membrane glycoprotein, which is synthesized with a signal peptide (aa 1-26) that is subsequently cleaved off to produce the mature protein that contains an extracellular domain (aa 27-332), a transmembrane domain (aa 333-355), and a cytoplasmic domain (aa 356-567). The full-length IL-33R contains three Ig-like C2 type motifs that are responsible for extracellular ligand-binding. It also contains a TIR domain (aa 380-540) that is critical for initiating intracellular signaling. The IL-33/IL-33R axis plays a vital role in both innate and adaptive immunity and is involved in the development of multiple types of immune-related diseases. Two isoforms of IL-33R are described that are produced by alternative splicing. The soluble form (sST2) serves as a decoy receptor and lacks the transmembrane and cytoplasmic domains. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonal. Physical form

Purified recombinant rat monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
