Specificity

Clone 2I13 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects CD48. It targets an epitope within 24 amino acids from the C-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 24 amino acids from the C-terminal half of mouse CD48.

Application

Quality Control Testing

Evaluated by Western Blotting in EL4 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected CD48 in EL4 cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected CD48 in lysates from RAW264.7 cells and Mouse thymus tissue.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected CD48 in Mouse spleen tissue sections.

Flow Cytometry Analysis: 1 µg from a representative lot detected CD48 in one million C57BL6 Mouse Splenocytes .

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected CD48 in RAW264.7 cells.

Affinity Binding Assay Analysis: A representative lot of this antibody bound CD48 peptide with a KD of 2.6 x 10-8 in an affinity binding assay.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

CD48 antigen (UniProt: P18181; also known as BCM1 surface antigen, BLAST-1, HM48-1, MRC OX-45 surface antigen, SLAM family member 2, SLAMF2, Signaling lymphocytic activation molecule 2, sgp-60, CD48) is encoded by the Cd48 (also known as Bcm-1) gene (Gene ID: 12506) in murine species. CD48 is a GPI-anchored cell surface glycoprotein that is synthesized with a signal peptide (aa 1-22) and a propeptide (aa 218-240 that are subsequently cleaved off to produce the mature protein. It contains two Ig-like C2 type domains (aa 29-125 and 129-209). CD48 exists in both a membrane-bound and soluble form. It interacts via its N-terminal immunoglobulin domain with cell surface receptors including CD2 to regulate immune cell function and activation. It participates in T-cell signaling transduction by associating with CD2 and efficiently bringing the Src family protein kinase LCK and LAT to the TCR/CD3 complex. In turn that promotes LCK phosphorylation and subsequent activation. It induces the phosphorylation of the cytoplasmic immunoreceptor tyrosine switch motifs (ITSMs) of CD244 initiating a series of signaling events that leads to the generation of the immunological synapse and the directed release of cytolytic granules containing perforin and granzymes by T-lymphocytes and NK-cells. CD48 expression is observed in mature lymphocytes and monocytes, but not on non-hematopoietic cells. Its expression is increased under inflammatory conditions. It is reported to be constitutively expressed at high levels on almost all multiple myeloma cells. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
