Specificity

Clone 1N9 is a ZooMAb® recombinant rabbit monoclonal antibody that specifically detects TAOK2. It targets an epitope within 25 amino acids from the N-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 25 amino acids from the N-terminal half of human TAOK2.

Application

Quality Control Testing

Evaluated by Western Blotting in MCF7 cell lysate.

Western Blotting Analysis (WB): A 1:10,000 dilution of this antibody detected TAOK2 in MCF7 cell lysate.

Tested Applications

Affinity Binding Assay: A representative lot of this antibody bound TAOK2 peptide with at least ten thousand-fold (10,000X) higher than with non-specific control peptide.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected TAOK2 in human liver tissue sections and in human cerebral cortex tissue sections.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Serine/threonine-protein kinase TAO2 (UniProt: Q9UL54; also known as EC:2.7.11.1, Kinase from chicken homolog C, hKFC-C, Prostate-derived sterile 20-like kinase 1, PSK-1, PSK1, Prostate-derived STE20-like kinase 1, Thousand and one amino acid protein kinase 2) is encoded by the TAOK2 (also known as KIAA0881, MAP3K17, PSK, PSK1) gene (Gene ID: 9344) in human. The encoded Kinase belongs to the MAP Kinase Kinase Kinase (MAPKKK) family of proteins. The gene is mapped to human chromosome 16p11.2, a region implicated in susceptibility to autism spectrum disorders and schizophrenia. The encoded protein modulates gene transcription by activating mitogen-activated protein (MAP) kinase pathways, such as JNK, p38, or extracellular signal-regulated kinase. It functions in the regulation of the actin cytoskeleton in non-neuronal cells through the activation of JNK. Its a multi-pass membrane protein and contains a conserved Protein kinase domain (aa 28-281) at the N-terminus and three coiled-coil domains. TAOK2 is ubiquitously expressed, with a higher level of expression in testis and brain. It primarily localizes in the cytoplasm but also found in the nucleus and associated with the Golgi apparatus. In neurons, TAOK2 (isoform 2) is localized in dendrites and synapses. Four isoforms of TAOK2 are known to be produced by Alternative splicing. Down-regulation of TAOK2 inhibits the formation of basal dendrites and axonal elongation. Therefore, it plays an important role in basal dendrite formation in the neocortex. Mutations or dysregulation of TAOK2 is linked to autism spectrum disorders (ASD) and intellectual disabilities. TAOK2 is also known to be associated Spondylocostal dysostosis 5 disease and may play a role in neurodegenerative conditions. This ZooMAb® recombinant monoclonal antibody, generated by our proprietary technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
