Specificity

Clone 1K16 is a ZooMAb® Rabbit recombinant monoclonal antibody that detects Inter- a-trypsin inhibitor heavy chain H1 (ITIH1/SHAP). It targets an epitope within 20 amino acids from the C-terminal region of mature ITIH1/SHAP.

Immunogen

KLH-conjugated linear peptide corresponding to 20 amino acids the C-terminal region of human mature ITIH1/SHAP (without signal peptide and propeptides).

Application

Quality Control Testing

Evaluated by Western Blotting in Human serum.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected ITIH1 in Human serum.

Tested applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected ITIH1 in Human plasma.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected ITIH1 in human placenta tissue sections.

Target description

Inter-alpha-trypsin inhibitor heavy chain H1 (UniProt: P19827; also known as ITI heavy chain H1, ITI-HC1, Inter-alpha-inhibitor heavy chain 1, Inter-alpha-trypsin inhibitor complex component III, Serum-derived hyaluronan-associated protein, SHAP) is encoded by the ITIH1 (also known as IGHEP1) gene (Gene ID: 3697) in human. The inter- a-trypsin inhibitor (ITI) family is a group of proteins that are composed of variable associations of light (ITIL) and five heavy chains (ITIH1, -H2, -H3, -H4, -H5), encoded by different genes. They contribute to extracellular matrix stability by covalent linkage to hyaluronan. Heavy chains represent the main structural unit of the ITI complex. The heavy chains are linked to light chain via a single chondroitin sulfate chain, which makes ITI a both structurally and functionally unique proteoglycan with a plasma protease inhibitory activity that resides solely in the light chain. ITIH1 is a glycoprotein that is synthesized in the liver and released into circulation. It is synthesized with a signal peptide (aa 1-27) and two pro-peptides (aa 28-34 and 673-911) that are subsequently cleaved off to generate the mature form (aa 35-672). ITIH1 serves as a carrier of hyaluronan in serum or acts as a binding protein between hyaluronan and other matrix protein, including those on cell surfaces in tissues to regulate the localization, synthesis, and degradation of hyaluronan. ITIH1 is suggested to have an antitumor role. It is shown to increase cell attachment and reduce the number of metastases. Down-regulation of ITIH genes have been reported in multiple solid tumors, including breast, colon, and lung cancer. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
