Rat glioma x rat glioma hybrid

Cell Line Description

Pseudohexadecaploid cell line generated by fusion of the 8-bromodeoxyuridine resistant mutant rat octaploid glioma cell line C6-3-BU-8 and the 6-thioguanine-resistant mutant rat octaploid glioma cell line C6-3-TG-3. C6-4-2 cells provide a suitable model system for studying the regulation of the membrane potential by peptide hormones.

Culture Medium

DMEM + 2 mM glutamine + 10% Foetal Bovine Serum (FBS) + HAT (0.1 mM hypoxanthine, 10 μM aminopterin and 16 μM thymidine) Cells can be cultured without HAT provided they were cultured in between in HT medium for 3-4 days

Subculture Routine

Split sub-confluent cultures (70-80%) 1:3 to 1:6 i.e. seeding at 2-4x10,000 cells/cm2 using 0.05% trypsin/EDTA; 5% CO2; 37°C
