General description

26S proteasome non-ATPase regulatory subunit 14 is also known as 26S proteasome regulatory subunit RPN11 or 26S proteasome-associated PAD1 homolog 1. This protein is expressed from the PSMD14 gene and is the metalloprotease component of the 26S proteasome that specifically cleaves ′Lys-63′-linked polyubiquitin chains. 26S proteasome non-ATPase regulatory subunit 14 has a role in the response to double-strand breaks (DSBs) by regulating non-homologous end joining (NHEJ) via cleaving ′Lys-63′-linked polyubiquitin, retaining JMJD2A/KDM4A on chromatin and restricting TP53BP1 accumulation. 26S proteasome non-ATPase regulatory subunit 14 is also involved in homologous recombination repair by promoting RAD51 loading. 26S proteasome non-ATPase regulatory subunit 14 is widely expressed, but found in the highest levels in heart and skeletal muscle.
