General description

Anti-Muellerian hormone type-2 receptor (EC 2.7.11.30; UniProt Q16671; also known as Anti-Muellerian hormone type II receptor, AMH type II receptor, MIS type II receptor, MISRII, MRII) is encoded by the AMHR2 (also known as AMHR, MISR2) gene (Gene ID 269) in human. Anti-Müllerian hormone (AMH) or Müllerian inhibiting substance (MIS) is a TGF- family homodimeric glycoprotein hormone. AMH/MIS is responsible for regression of the Müllerian ducts in male embryos, it is also produced in both male and female gonads after birth and plays a role in folliculogenesis, germ cell maturation and gonadal function. AMH/MIS interacts with single membrane-spanning serine/threonine kinase receptors type I (AMHR1/MISRI) and II (AMHR2/MISRII). AMHR1 is nearly ubiquitously expressed, whereas AMHR2 is mainly detected in the gonads and other organs of the reproductive tract. AMHR2 is found expressed in the majority of human granulosa cell tumors (GCTs) and epithelial ovarian cancers (EOCs). In addition, AMHR2 expression is also reported in breast and prostate cancers. Human AMHR2 is produced with a signal peptide (a.a. 1-17), which is removed posttranslationally to yield the mature receptor with an N-terminal extracellular region (a.a. 18-149), a singl-transmembrane domain (a.a. 150-170), and a large cytoplasmic region (a.a. 171-573) that contains the kinase domain (a.a. 203-518; a.a. 230 ATP-binding site; a.a. 333 proton acceptor site; a.a. 209-217 ATP-binding site).

Specificity

Clone 10H8.1 targets an extracellular epitope present in all three spliced isoforms of human AMH type II receptor (AMHR2) reported by UniProt (Q16671).

Immunogen

GST-tagged recombinant human AMHR2 extracellular domain.

Application

Detect AMHR2 using this mouse monoclonal Anti-AMHR2 Antibody, clone 10H8.1, Cat. No. MABC1213, validated for use in Western Blotting.

Quality

Evaluated by Western Blotting in Jurkat cell lysate.

Western Blotting Analysis: 0.5 µg/mL of this antibody detected AMHR2 in 10 µg of Jurkat cell lysate.

Target description

~60 kDa observed. 62.75/52.24/51.83 kDa (isoform 1/2/3) calculated. Uncharacterized band(s) may appear in some lysates.
