General description

Histone H3 is one of the five main histone proteins involved in the structure of chromatin in eukaryotic cells. Featuring a main globular domain and a long N-terminal tail, H3 is involved with the structure of the nucleosomes of the ′beads on a string′ structure. Histone proteins are highly post-translationally modified, and Histone H3 is the most extensively modified of the five histones. The term "Histone H3" alone is purposely ambiguous in that it does not distinguish between sequence variants or modification state. Histone H3 is an important protein in the emerging field of epigenetics, where its sequence variants and variable modification states are thought to play a role in the dynamic and long term regulation of genes.

Specificity

Broad species cross-reactivity is expected, based on sequence homology.
This antibody recognizes Histone H3 that is unmodified at Thr3 and Lys4 but does not recognize phosphorylated Thr3 nor acetylated or methylated Lys4. The antibody binding specificity allows modifications of Lys9, Ser10, and/or Lys141.

Epitope: N-terminus (unmodified)
Synthetic peptide corresponding to amino acids 1-12 of human Histone H3, conjugated to KLH.

Quality

Routinely evaluated by Western Blot on HeLa cell lysate.

Western Blot Analysis: 1:10,000 dilution of this antibody detected histone H3 in 10 μg of HeLa Acid Extract cell lysate and recombinant histone H3 (500 ng) but not other recombinant histones.

Storage and Stability

Stable for 1 year at 2-8°C from date of receipt.
For maximum recovery of product, centrifuge the vial prior to removing the cap. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
