Specificity

Clone 4K14 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects DDX5. It targets an epitope within 19 amino acids from the C-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 19 amino acids from the C-terminal half of human Probable ATP-dependent RNA helicase DDX5.

Application

Quality Control Testing

Evaluated by Western Blotting in SH-SY5Y cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected DDX5 in SH-SY5Y cell lysate.

Tested applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected DDX5 in C6, NIH3T3, Cos7, and U2OS cell lysates.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected DDX5 in human cerebral cortex tissue sections.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected DDX5 in SH-SY5Y cells.

Affinity Binding Assay: A representative lot of this antibody bound DDX5 with a KD of 6.3 x 10-9 in an affinity binding assay.

Target description

Probable ATP-dependent RNA helicase DDX5 (UniProt: P17844; also known as EC:3.6.4.13, DEAD box protein 5, DDX5, RNA helicase p68) is encoded by the DDX5 (also known as G17P1, HELR, HLR1) gene (Gene ID: 1655) in human. DDX5 is a member of the DEAD box protein family that is characterized by a conserved Asp-Glu-Ala-Asp (DEAD; aa 248-251) motif within a helicase core. It contains a helicase ATP-binding domain (aa 125-300) and a helicase C-terminal domain (aa 328-475). It is present in cytoplasm and in the nucleus. During the G0 phase, it is predominantly located in the nucleus. Its cytoplasmic levels increase during the G1/S phase and during the M phase it is located in the vicinity of the condensed chromosomes. DDX5 is an ATP-dependent RNA helicase that participates in unwinding of RNA secondary structure, RNA splicing, mRNA export, transcript stability, rRNA biogenesis, and microRNA processing. It is also involved in transcriptional regulation and this function is independent of its RNA helicase activity. It also serves as a transcriptional coactivator for p53/TP53 and involved in p53/TP53 transcriptional response to DNA damage and p53/TP53-dependent apoptosis. DDX5 is essential for regulation of appropriate splicing of key genes necessary for spermatogenesis. It regulates expression of cell cycle genes in undifferentiated spermatogonia post-transcriptionally and is required for cell proliferation and survival. Overexpression of DDX5 has been reported in several human cancers and is implicated in tumorigenesis. It activates the Myc expression in multiple solid tumors. It is reported to preferentially unfold the Myc GQ structure to activate Myc expression in several cancer cell lines. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
