General description

Exosome component 8 (EXOSC8) or RRP43, a 3′ to 5′ exoribonuclease, is a component of the eukaryotic exosome (a 400 kDa complex). EXOSC8 is part of the hexameric ring, along with EXOSC4-7 and EXOSC9. This is further capped by three RNA binding subunits (EXOSC1-EXOSC3). EXOSC8 gene is mapped to human chromosome 13q13.3 and is localized in nucleus and cytoplasm.

Specificity

Anti-EXOSC8 (N-terminal) recognizes human, mouse and rat EXOSC8.

Immunogen

peptide corresponding to the N-terminal region of human EXOSC8, conjugated to KLH.

Application

Anti-EXOSC8 (N-terminal) antibody produced in rabbit may be used in immunoblotting and immunoprecipitation.

Biochem/physiol Actions

Exosome component 8 (EXOSC8) interacts with AU-rich element (ARE)-containing mRNAs and its deficiency halts growth in yeast. Mutations in the EXOSC8 gene is implicated in Ponto-cerebellar hypoplasia.

Physical form

Solution in 0.01 M phosphate buffered saline pH 7.4, containing 15 mM sodium azide.

Storage and Stability

For continuous use, store at 2-8°C for up to one month. For extended storage freeze in working aliquots. Repeated freezing and thawing is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilution samples should be discarded if not used within 12 hours.
