General description

p53 is a tumor suppressor gene expressed in a wide variety of tissues. It is a tetrameric nuclear DNA-binding phosphoprotein. The gene encoding it is localized on human chromosome 17p13.1 and mouse chromosome 11.

Immunogen

recombinant human wild type p53. The epitope resides beween amino acids 20-25 of human p53.

Application

Monoclonal Anti-p53-Biotin antibody produced in mouse has been used for Western Blotting.

Biochem/physiol Actions

Tumor suppressor p53 has the capability to induce cell cycle arrest and has a role in DNA repair, senescence and apoptosis. It binds to Simian vacuolating virus 40 (SV40) T-antigen and human papilloma virus E6 protein. The p53 gene is mutated in various cancers, such as of the breast, ovarian, bladder, colon and lung.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 1% bovine serum albumin and 15 mM sodium azide.

Storage and Stability

For continuous use, store at 2-8°C for up to one month.
For extended storage, freeze in working aliquots.
Repeated freezing and thawing, or storage in “frostfree”freezers, is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilution samples should be discarded if not used within 12 hours.

Other Notes

In order to obtain the best results using various techniques and preparations, we recommend determining optimal working dilutions by titration.
