General description

Monoclonal Anti-Rabbit Immunoglobulins (mouse IgG1 isotype) is derived from the hybridoma produced by the fusion of mouse myeloma cells and splenocytes from an immunized mouse. Immunoglobulins is composed of two heavy (H) and two light (L) chains. Each containing one NH2-terminal variable region and one or more COOH-terminal constant regions. They are heterodimeric proteins.
Reacts with an epitope on the heavy chain of rabbit IgG, IgA and IgM, which is sensitive to reduction. No cross-reactivity with human IgG, IgA and IgM.

Specificity

Binds only rabbit Igs. Does not react with reduced rabbit Igs.
No cross reaction is observed with IgG from the following species: bovine, cat, chicken, dog, goat, guinea pig, horse, pig, rat, sheep or turkey.

Application

Monoclonal Anti-Rabbit Immunoglobulins Alkaline Phosphatase antibody produced in mouse has been used in:
  • indirect enzyme linked immunosorbent assay (ELISA)
  • immunoblotting
  • immunohistochemistry

The mouse monoclonal anti-Rabbit IgG (heavy chain-specific) peroxidase conjugated antibody can be used as a secondary antibody for western blot analysis of protein preps from E. coli cultures at a concentration of 1:10000 diluted in PBSt for 1 hour at room temperature.

Biochem/physiol Actions

IgGs are glycoprotein antibodies that modulate several immune responses. Rabbit IgGs against target proteins are often used as primary antibodies in various research applications. Thus, secondary anti-rabbit IgGs conjugated to a detectable substrate are useful tools for the analysis of target proteins.
Mouse monoclonal anti-rabbit immunoglobulins-alkaline phosphatase antibody binds to an epitope on the heavy chain of rabbit IgG, IgA, and IgM. The product stains bands at the intact whole molecule and at the heavy chains in an immunoblot of denatured, non-reduced rabbit immunoglobulins. Reduction of rabbit immunoglobulins destroys the epitope. In ELISA, the product shows no cross reaction with human serum or tissue preparations, nor with human IgA, IgG, or IgM.
The constant domains of immunoglobulins are involved in the complement activation.

Physical form

Solution in 0.05 M Tris buffer pH 8.0, containing 1 mM MgCl2, 0.1% bovine serum albumin, 50% glycerol and 15 mM sodium azide
