General description

Nicotinamide (NAM) belongs to vitamin B family and is the amide form of vitamin B3. NAM is present in food and is a constituent of nicotinamide adenine dinucleotide (NAD) and nicotinamide adenine dinucleotide phosphate (NADP). Tryptophan is the precursor for NAM synthesis in liver.[1] NAM controls cell metabolism, redox reactions, mitochondrial functionsand energy generation. It promotes natural killer cells proliferation and survival.[2] NAM confers neuroprotection and promotes neuronal maturation. It has the ability to cross the blood-brain barrier and may provide protection in ischaemic stroke and traumatic brain injury. NAM may elicit therapeutic benefits in Alzheimer′s, Parkinson′s, and Huntington′s diseases.[3]

Application

Nicotinamide has been used as a medium supplement in organoid culture of intestinal stem cells[4], esophageal squamous cell carcinomas(ESCC)[5], pancreatic cancer organoids[6] and pancreatic progenitor induction.[7]

    1. The recommended storage conditions and stability for stock solutions, once prepared, are stored aliquoted in tightly sealed vials at -20°C or below and used within 1 month. Wherever possible, solutions should be made and used on the same day.

    1. The product in solution has not been tested for stability following multiple freeze/thaw cycles. As a general practice, multiple freeze/thaw cycles are avoided.

  4. How long are aliquots of a 1 M solution in DPBS or water at °70°C stable?

    1. Solution stability studies at 70°C have not been performed on this product. Neutral aqueous solutions are fairly stable and may be autoclaved at 120 °C for up to 20 minutes. Solutions at an alkaline pH will be hydrolyzed to nicotinic acid. Nicotinamide is a component of several vitamin solutions used in cell culture (listed as niacinamide). These products are stored at -20°C, but are typically added to culture medium at 37°C.
