General description

Epo is mainly produced by liver cells in fetal stages, by kidneys under hypoxic conditions; liver and brain also secrete Epo in less amounts. Anti-Erythropoietin antibody specifically reacts with human Erythropoietin. The EPO gene is mapped to human chromosome 7q22.1.

Immunogen

recombinant human erythropoietin expressed in CHO cells.

Application

Anti- Erythropoietin antibody may be used for immunoblotting at a working concentration of 1 μg/mL. For immunohistochemistry, a working concentration of 5-15 μg/mL may be used. The antibody is also suitable for neutralization reactions. Erythropoietin antibody has been used for ELISA in the extracts of leaves of tobacco plant in which recombinant human erythropoietin was expressed. The antibody has also been used for immunoaffinity chromatography to separate the leaf extracts of tobacco expressing recombinant human erythropoietin.

Biochem/physiol Actions

Erythropoietin (Epo) is a pleotropic cytokine that promotes erythropoiesis or proliferation and differentiation of red blood cell precursors. The red blood cell production is stimulated when Epo binds and activates the Epo receptor (EpoR) dimer. EpoR directly regulates the signalling by Jak2, PI3K, NF-κB, STAT5A and STAT5B. Epo also inhibits apoptosis of erythroid progenitors by downregulating Fas (tumor necrosis factor receptor superfamily member 6) and FasL (Fas ligand). The expression of Epo is under the control of the transcription factor hypoxia inducible factor (HIF).
