Gene Information

mouse ... Junb(16477)

General description

Transcription factor Jun-B (UniProt: P09450; also known as MyD21) is encoded by the Junb (also known as Jun-b) gene (Gene ID: 16477) in mouse. JunB is a member of the AP-1 transcription family that binds to the DNA sequence 5′-TGA[CG]TCA-3′ and is involved in regulating gene activity following the primary growth factor response. It can bind to DNA either as a homodimer or as a heterodimer with another member of the Jun/Fos family. JunB plays an important role in proliferation, differentiation, and death. With respect to cell cycle control, JunB is reported to have a dual function. It can serve as a cell division inhibitor and a tumor suppressor or can promote cell division. It is reported to diminish cell proliferation via transcriptional activation of p16 (INK4a). Its expression is shown to be very low in quiescent cells but can be rapidly induced by mitogenic stimuli during the G0/G1 transition. It induces proliferation, survival, drug resistance in multiple myeloma (MM) cells and silencing of JunB is reported to block MM cell proliferation in a murine xenograft model. It also protects MM cells against dexamethosome and bortezomib-induced cell death. JunB is also reported to regulate epidermal stem cells and sebaceous glands through balancing proliferation and differentiation of progenitors and by suppressing lineage infidelity. Specificity

Clone 75-5 is a mouse monoclonal antibody that detects human JunB. It targets an epitope within the N-terminal region.

Immunogen

GST-tagged recombinant fragment correspond to the first 75 amino acids from the N-terminal region of mouse JunB.

Application

Quality Control Testing

Evaluated by Western Blotting in HeLa cell lysates.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected JunB in HeLa cell lysates.

Tested Applications

Western Blotting Analysis: A representative lot detected JunB in Western Blotting applications (Passegue, E., et. al. (2000). EMBO J. 19(12):2969-79; Farras, R., et. al. (2008). Mol Cell Biol. 28(12):4173-87; Gomard, T., et. al. (2010). PLoS One. 5(3):e9585).

Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected JunB in HeLa cell line.

Immunocytochemistry Analysis: A representative lot detected JunB in Immunocytochemistry applications (Bakiri, L., et. al. (2000). EMBO J. 19(9):2056-68).

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user
